Acquity Sample Loop Change
I'm looking to increase my sample injection volume for a dissolution application where the samples will be very dilute. We have a recently purchased Acquity (old, used, but functional) with sample manager running on Empower 3, and I'm unsure how large I can go with injection volume. The sample manager is a 186015006, currently has a 5uL loop with 100uL sample syringe. I've gathered I can go to a 50uL loop (430001325), for which I think I need to step up to a 250uL syringe (410001347). However in the configuration portion of the software, there is option for an even larger 250 uL loop, but I'm unsure if the hardware is compatible? I think I would need a 500 uL sample syring for such a loop. Coming from Thermo & Agilent background so I'm struggling a bit with the switch and would appreciate any help. Happy to read a manual, but to be honest I don't know which one applies to this system as it seems there are many different versions of an Acquity over the years.

Hi MitohH,
The largest injection that you can make on that autosampler is 250uL (sample loop PN 430002007). This can be done with the 100uL syringe which is the largest available. To accomplish this, the syringe will have to do multiple draws from the sample vial which increases the injection volume and reproducibility error. Also keep in mind that the instrument has multiple injection modes with differing volume requirements (usually more than the programed injection volume) so vial capacity may also be a consideration.
Good luck!

Thank you @MarcNoble, I appreciate the help. Is there a hardware manual that I can reference? The closest I have found is a 2006 version of the Acquity UPLC Operators Guide (71500082502 / Revision C), but it only mentions a 50uL fixed loop as an option, and says that requires a 250 uL syringe. I wasn't sure if that's the right manual to reference though. As I understand it, I'm not working with an H-Class and don't have FTN, just standard fixed loop Acquity UPLC?
It appears that is an expansion loop you gave the part number for; is that installed between the 6-port and the needle, leaving the 5uL fixed loop in place?
Understood on vial capacity, currently configured for 2mL vials.0 -
Update on this: Classic Acquity, fixed loop (non FTN), need a 500 uL syringe (410001647) with 250 uL loop (430002211), which is available in kit form as 176001780.0
