HILIC - system conditions
<p>Hi,</p><p></p><p>We're developing a method using an Acquity UPLC BEH HILIC column - it's the first time I've used this separation technique, much more comfortable with reverse phase so it's hurting my brain a little!</p><p></p><p>Our initial gradient starts at 95% MeCN/5% buffer and our analyte is eluting where we want, but the peak shape is poor. I'm guessing this could be caused by the wash solutions or the sample diluent.</p><p></p><p>At the moment sample diluent is 80%MeCN/20% H2O and both strong and weak needle washes are 75%MeCN/25%MeOH.</p><p></p><p>I think the weak needle wash is OK (according to the column care notes), but can anyone suggest any changes I could make to improve the chromatography.</p><p></p><p>Thanks</p>
