Linear response

<p>Why am I not getting a linear response for a serial dilution of 1, 10 100 ugs/ml solutions at 214 nm on the Acquity TUV?  1 and 10 look good but 100 is about half of what it should be.  The chromatograms look fine and the peaks are symetrical and well integrated.Thanks.</p>

    Hi Susan

    I assume that you are injecting the same volume of all 3 dilutions ? What are the absorbance values (AU) for your 3 peaks ? Are you working inside the linear area of the detector ? Some chromatograms might help troubleshoot as well as details of the conditions, instrument settings and mobilephase etc.
