Phase 2 Acquity Column Issues

<p><span>Has anyone noticed a difference in performance with the new Phase 2 Acquity columns? For some applications we have experienced a drop in number of samples that can be chromatographed when compared with the same application on the "Phase 1" columns. In particular, it appears that either a column void has developed or that the column frit has become contaminated or maybe both. In etiher case, peak shape drops off much more rapidly than with the older columns. While we have not completely eliminated our methodology as the source of problem, the drop in performance has caused lingering doubts about UPLC robustness to resurface. </span></p>

  • Hi Daniel - I have not seen a drop off in performance with the Phase II columns. The resolution capapbiltes seem to be the same as well. My problem is because of the formulation I am assaying - my column life time is about 300 to 400 injections -max - regardless of Phase I or II columns. Do you filter mobile phase and are you using phosphate buffers?

    I am sorry to hear of your UPLC column problems. I am hoping that some users can chime in with additional UPLC column lifetime comments.

    Generally speaking, we've not seen much difference (Phase I vs II) in the very low number of UPLC column compliants. That said, I would like to see if we can fix your column problems sooner rather than later, assuming the problem IS the column(s). One way to do that is to get a new column into your hands.

    Have you been in touch with your local Chemistry Specialist? If not, I would be more than happy to ask that person to get in touch with you. The reason I ask this is that I would like to get a new column to you as soon as possible and your Chemistry Specialist can facilitate this for you (or I can help if you prefer).

  • Yes. I have had similar problems. I am injecting neat reaction mixtures (filtered) and had outstanding lifetimes with the version 1 columns. We noticed a substantial drop in the number of injections we were getting on the version 2 columns. Waters is working with us to try to resolve the issue. In the meantime, I am trying a gradual ramping of the pressure (going from 0.1 mL/min to 1mL/min) instead of the instant 0-1mL/min to hopefully lessen the blow the pressure causes to the inlet of the column.
