Updating Ion Ratios

<p><span>Hello</span></p><p></p><p><span>I am trying to figure out how to make my method automatically update ion ratios using MassLynx (QuanLynx) V4.1. If I click on "Update Ion Ratios" it will update the ion ratios from my first standard but since my first standard is 0 (zero) it obviously is not helpfull since it updates the ratio to zero since nothing is there.</span></p><p></p><p><span>The instructions say you can update using multiple reference samples using the QUAN_REF field in the MassLynx sample list. Instructions are to "make an entry in the QUAN_REF field for each of the reference samples." Well what is the "entry?" There is no drop down menu associated with that field.</span></p><p></p><p><span>TIA</span></p><p><span>Mike</span></p>

  • I'm not 100% sure, but I think you put an * in that column for whatever injections you want to use to update it. And then in the processing method, you have to make sure the update ion ratios option is set to yes. :)
  • I called technical support and Kathy told me to try an X and it worked. After getting your response I tryed * and that also works.

  • Highlight a line in your sample list that is a standard that has appropriate peak ratios to work with, usualy I pick my low level control. From the QuanLynx or Target Lynx menu select process samples and make sure you have ticked the box for update ratios.

    Once in Quanlynx viewer go to the file menu and select export method to save a new copy of the method with updated ratios.

    Next time you process make sure you have unchecked the update ratios box before processing any data. Double check that the ratios are sustainable or you won't see any results for your samples, I usually start with 20%.

    There are several ways to update ion ratios.

    1. choose a sample in the sample list by marking it with an X in the quan ref column before you process your sample list. advantage: this can be done on multiple samples (if you check this option in the method). this way the ion ratio will be updated using the average of all marked reference samples. drawback: when you mark the quan ref column with X before you process the data, you are never 100% sure you have chosen a good reference sample. Bear in mind that once you have marked X in the quan ref column, QuanLynx will always use these samples as references, even if you want to choose option 2 afterwards. If you want to use option 2 after you have marked X in the quan ref column, you will have to remove them first and then reprocess the sample list.

    2. do not use the quan ref column, but process your sample list and highlight one sample in our QuanLynx summary table and press processig > update ion ratios. this way the ion ratio will be updated using that one sample. the advantage is that you ca choose at any time which sample to use as reference sample. the drawback is that only one sample can be used as reference (no average of several samples like in the quan ref column).
